Usuthu are hoping to secure their first league win of the 2019/20 season having lost their first two matches.
While Pirates are targeting their second victory in the new campaign after losing to SuperSport United in their previous game in the competition.
Pirates are undefeated in their last five league matches against AmaZulu.
Bucs have recorded four victories and one draw against Usuthu.
Meanwhile, Mamelodi Sundowns will take on Cape Town City at the Lucas Moripe Stadium on Tuesday, August 20.
Sundowns remain undefeated in the league in the new season having drawn with Chippa United in their last match.
On the other hand, City are also undefeated in the league having defeated their Western Cape rivals Stellenbosch in their previous match.
City have dominated their last five league matches against Sundowns.
The Citizens have recorded three victories compared to the Brazilians' two wins.
Lastly, Kaizer Chiefs will lock horns with SuperSport United at the FNB Stadium on Saturday, August 24.
Chiefs have started the new season with two successive wins over Highlands Park and Black Leopards.
While SuperSport recorded a defeat to Sundowns in their first game, but they defeated Pirates in their second match.
The last five league matches between Chiefs and SuperSport have produced three draws.
The two teams have recorded one victory each.
